Acid and enzymatic hydrolysis to recover reducing sugars from cassava bagasse: an economic study BABT
Woiciechowski,Adenise Lorenci; Nitsche,Saul; Pandey,Ashok; Soccol,Carlos Ricardo.
The objective of this work was to study the acid and enzymatic hydrolysis of cassava bagasse for the recovery of reducing sugars and to establish the operational costs. A statistical program "Statistica", based on the surface response was used to optimize the recovery of reducing sugars in both the processes. The process economics was determined considering the values of reducing sugars obtained at laboratory scale, and the operations costs of a cylindrical reactor of 1500 L, with flat walls at the top and bottom. The reactor was operated with 150 kg of cassava bagasse and 1350 kg of water. The yield of the acid hydrolysis was 62.4 g of reducing sugars from 100 g of cassava bagasse containing 66% starch. It represented 94.5% of reducing sugar recovery. The...

Activity of fungal phytases stored in two ways in response to the period of storage at room temperature Ciência Rural
Naves,Luciana de Paula; Corrêa,Angelita Duarte; Bertechini,Antônio Gilberto; Gomide,Elisangela Minati; Abreu,Celeste Maria Patto de.
Two distinct experiments were conducted simultaneously with phytases of Aspergillus oryzae and A. niger for determining enzyme activity in response to storage period (up to 180 days at room temperature - RT). In the first experiment, enzymes were stored as they were acquired (pure form) and the activity was measured periodically during 180 days of storage at RT. In the second experiment, the phytases were incorporated to a supplement containing vitamins, minerals, and amino acids; and then this supplement was stored at RT up to 180 days, so that every 30 days of storage was collected one aliquot from each replicate for determining enzymatic activity. In conclusion, the phytase activity is affected by storage duration. To ensure 80% of the initial activity,...

Adaptation du métabolisme respiratoire de l'huître creuse Crassostrea gigas ArchiMer
Le Moullac, Gilles.
Summer mortality of the oyster result from the interaction of many factors. A respiratory assumption proposes a link between the supposed causes of the energy state, the cultivation methods and the medium. This work is focused towards the search for specific metabolic markers of environmental stresses. The cDNA of genes coding for PK and PEPCK were cloned and sequenced in order to detect their regulation level. The sediment nearness affects the state of energy reserves, the metabolic pathways of energy production implying a regulation of the PK. Face to the dissolved oxygen decline, oysters were oxyregulator but with a low regulation ability. In hypoxia, glycolysis was slowed down. PK activity was inhibited resulting from an increase in alanine sensitivity...

Assessment of biochemical methods to detect enzymatic depolymerization of polysaccharides ArchiMer
Rigouin, Coraline; Delbarre Ladrat, Christine; Sinquin, Corinne; Colliec-jouault, Sylvia; Dion, M.
To find biocatalyst allowing depolymerization of new polysaccharides, one needs to get a sensitive and well adapted method to a screening program. This led us to compare biochemical methods of detection of the depolymerization. Currently used methods such as reducing sugars assays, double bond monitoring or molecular weight determination were tested to follow the kinetic of depolymerization with different enzyme / polysaccharide couples. The range of concentrations of assorted enzymes allowed us to identify the most sensitive and appropriate method to detect polysaccharide degradation. Reducing sugars assays are quantitative, sensitive and almost usable with all kind of polysaccharide but some compounds may interfere with them. When polysaccharide is...

Attendrissement enzymatique de chair de mollusques ArchiMer
Kouadio Kouakou, François.
Two types of marine molluscs were studied. The highly individual characteristics of the flesh of these two shellfish, in this case its firmness, led us to analyse the behaviour of the foot muscle, the firmest part, as a function of heat and to the determination of the different temperatures at which the denaturation of the main constituent proteins occurs. This firmness makes the muscles difficult to chew to such an extent that they have acted as a break to consumption. These shell-fish have very similar proportions of myofibrillar and connective proteins, both of- which are responsible for the texture of meat. This similarity of composition determined the choice of an enzymatic preparation (protease V100) capable of acting simultaneously on both these...

Bioremediation of polyaromatic hydrocarbons (PAHs) using rhizosphere technology BJM
Bisht,Sandeep; Pandey,Piyush; Bhargava,Bhavya; Sharma,Shivesh; Kumar,Vivek; Sharma,Krishan D..
The remediation of polluted sites has become a priority for society because of increase in quality of life standards and the awareness of environmental issues. Over the past few decades there has been avid interest in developing in situ strategies for remediation of environmental contaminants, because of the high economic cost of physicochemical strategies, the biological tools for remediation of these persistent pollutants is the better option. Major foci have been considered on persistent organic chemicals i.e.polyaromatic hydrocarbons (PAHs) due to their ubiquitous occurrence, recalcitrance, bioaccumulation potential and carcinogenic activity. Rhizoremediation, a specific type of phytoremediation that involves both plants and their associated...

Carbohydrate metabolism of Xylella fastidiosa: Detection of glycolytic and pentose phosphate pathway enzymes and cloning and expression of the enolase gene Genet. Mol. Biol.
Facincani,Agda Paula; Ferro,Jesus Aparecido; Pizauro Jr.,João Martins; Pereira Jr.,Haroldo Alves; Lemos,Eliana Gertrudes de Macedo; Prado,Alessandro Luis do; Ferro,Maria Inês T..
The objective of this work was to assess the functionality of the glycolytic pathways in the bacterium Xylella fastidiosa. To this effect, the enzymes phosphoglucose isomerase, aldolase, glyceraldehyde-3-phosphate dehydrogenase and pyruvate kinase of the glycolytic pathway, and glucose 6-phosphate dehydrogenase of the Entner-Doudoroff pathway were studied, followed by cloning and expression studies of the enolase gene and determination of its activity. These studies showed that X. fastidiosa does not use the glycolytic pathway to metabolize carbohydrates, which explains the increased duplication time of this phytopatogen. Recombinant enolase was expressed as inclusion bodies and solubilized with urea (most efficient extractor), Triton X-100, and TCA....

Chicken serologic response to Salmonella enterica serotype Typhimurium assessed by Elisa Rev. Bras. Ciênc. Avic.
Oliveira,GH; Berchieri Jr,A; Montassier,HJ.
This study evaluated two enzyme-linked immunosorbent assays (ELISA) in the detection of chicken serologic response against Salmonella enterica sorotype Typhimurium. The assays have used as detecting antigen the soluble bacterial proteins of a non-flagellated strain of Salmonella Typhimurium (AgTM), and antibody conjugated to peroxidase or alkaline phosphatase. According to the results, optimal dilutions of antigen (concentration 5.49 mg/mL) and serum samples in both assays were 1:20,000 and 1:1,000, respectively. In such conditions, the ELISA/AgTM was able to detect serological response to Salmonella Typhimurium. Cross-reactions to Salmonella serotypes Gallinarum and Pullorum were seen, but not with other serotypes such as Enteritidis.

Comparative study on production of α-Amylase from Bacillus licheniformis strains BJM
Divakaran,Dibu; Chandran,Aswathi; Pratap Chandran,R.
Alpha amylase (α-1, 4-glucan-glucanhydrolase, EC 3.2.1.1), an extracellular enzyme, degrades α, 1-4 glucosidic linkages of starch and related substrates in an endo-fashion producing oligosaccharides including maltose, glucose and alpha limit dextrin (7). The present study deals with the production and comparative study of production of α-amylase from two strains of Bacillus licheniformis, MTCC 2617 and 2618, by using four different substrates, starch, rice, wheat and ragi powder as carbon source by submerged fermentation. The effect of varying pH and incubation temperature, activator, inhibitor, and substrate concentration was investigated on the activity of α-amylase produced by MTCC strain 2618. The results shows that the production of the α-amylase by...

Comparison of growth, digestive system maturation and skeletal development in sea bass larvae reared in an intensive or a mesocosm system ArchiMer
Zouiten, Dora; Ben Khemis, Ines; Masmoudi, Ahmed Slaheddin; Huelvan, Christine; Cahu, Chantal.
The quality of development in intensive or mesocosm hatchery-reared Dicentrarchus labrax larvae was investigated using physiological indicators assessing ontogeny. Larvae were reared in intensive (120 L tanks) and in mesocosm systems (20 m(3) enclosures) with the same feeding sequence, excluding the wild zooplankton naturally available in mesocosms. Faster growth was recorded since early development [16 day after hatching (DAH)] in the mesocosm. Maturation of the digestive system also occurred earlier as indicated by the higher amylase secretion ratios, the intestinal maturation index (alkaline phosphatase/leucine-alanine peptidase and aminopeptidase-N/leucine-alanine peptidase ratios) and the more developed intestinal epithelium at 23 DAH. Nevertheless,...

Comparisons Between Different Methods in Measuring Enzyme Similarity for Metabolic Network Alignment BABT
Zhou,Wenwei; Liu,Jiangtao; Xing,Hiu; Zhang,Zhengdong; Xie,Xiaoyao; Jing,Fengxuan.
Metabolic network alignments enable comparison of the similarities and differences between pathways in two metabolic networks and help to uncover the conserved sub-blocks therein. Such analysis is important in the understanding of metabolic networks and species evolution. The fundamental parts of metabolic network alignment algorithms all involve comparisons of the similarity between two enzymes as a similarity measure of network nodes. As a result, the study of methods for measuring enzyme similarity becomes highly relevant. Currently, two approaches are mainly used to measure enzyme similarity. One of the methods is based on similarity measures of gene or protein sequences; the other is based on enzyme classification. In this study, multiple metabolic...

Co-production of Proteases and Bioactive Protein Hydrolysates from Bioprocessing of Feather Meal BABT
Bernardo,Bernardete da Silva; Ramos,Rodrigo Ferraz; Callegaro,Kelly; Daroit,Daniel Joner.
Abstract: Feather meal conversion through submerged cultivations with Bacillus strains (CL33A, CL14) yielded proteases and protein hydrolysates. After 4-day (CL33A) and 10-day (CL14) cultivations, protease activities reached 461 U/mL; hydrolysates presented antioxidant (radicals-scavenging, 57-77%; Fe2+-chelation, 14-28%; Fe3+-reduction) and antidiabetic (dipeptidyl peptidase-IV inhibition, 49-52%) potentials. The obtained bioproducts present prospective commercial/industrial applications.

De l'utilisation de protéases industrielles pour la production de sauces de poisson ArchiMer
Le, Minh Chau.
Fish sauces are traditionally used in most Asian countries. Whatever the process, the principle is the same i.e. a fish autolysis in saline media followed by a fermentation step. Not less than 18 months are needed to obtain a brown liquid rich in salt and amino acids. This PhD work aims to determine if an addition of proteases can speed up the process while keeping the nutritional and sensorial properties. The first chapter presents the characteristics of fish sauces and the key parameters of the processes used. In the second chapter, several industrial proteases are compared each other for their capacity to liquefy the fishes in hyper saline conditions. Kinetics of proteolysis and biochemical characteristics of the resulting hydrolysates are established...

Dhal Recovery From Enzyme Pretreated Pigeon Pea Cultivar Gjp1 CIGR Journal
Dabhi, Mukesh N.; Sangani, V P; Rathod, P J.
GJP1 variety is newly developed variety of pigeon pea. It gives higher seed yield as compared to other varieties. Pigeon pea is mostly consumed in the form of splits hence, it is necessary to verify its hulling efficiency and utmost dhal recovery. Pre milling treatments are generally employed to loosen the seed coat to remove husk without losing any edible portion and better dhal recovery.  Xylanase, pectinase and cellulose enzymes were used to evaluate the milling properties of pigeon pea grains. Efforts were made to evaluate the effect of enzymatic parameters, i.e., enzyme concentration (20-50 mg/100g dry matter), incubation time (4-12 h), incubation temperature (35-55 oC) and tempering water pH (4.0-6.0) on hulling efficiency were optimized using...
